Apical membrane NaHCO3 cotransport has been demonstrated in the frog RPE.3738 Transepithelial HCO3 transport, in addition to Cl~, also is considered to drive fluid transport across the RPE.39"41 Because NaHCOs cotransport is electrogenic, with a HCO3 Na + ratio of between 1 and 3, 3738 it was conjectured that the light-evoked subretinal [K + ]o decrease, by hyperpolarizing the RPE, would reduce the rate of HCO3 influx across the apical membrane.37 This could contribute to the light-evoked increase in subretinal volume. However, the effect of NaHCO3 cotransport may be more complex and difficult to predict than this. For instance, a decrease in HCO3 influx will acidify he cell. Lin et al42 have shown that intracellular acidification stimulates Cl~ influx, and this would have the effect of decreasing subretinal volume. In the present experiments, we attempted to suppress RPE NaHCO; j cotransport by perfusing the retinal side of the preparation with DIDS, an inhibitor of the NaHCO3 cotransporter, 37 but this also affected the neural retina, and the results could not be interpreted. Huang and Karwoski9 showed that amiloride, which blocks Na + -H + exchange, significantly reduces the [TMA + ]O response. However, amiloride will change intracellular pH as well, and this can have consequences for Na-K-Cl cotransport at the apical membrane.CS 1 RPE Na-K-Cl cotransport can be modulated by changes in the concentration of any of the three ions, Na + , K + , Cl~. We reasoned that by changing ion concentrations in the bath facing the neural retina we would, by diffusion, also change its concentration in the SRS and affect RPE Na-K-Cl cotransport. These experiments were more successful for [Cl~]o changes than for [K + ]o. By adjusting the size and duration of low [Cl~] exposure in the retinal bath, we produced a subretinal [TMA + ]O decrease that resembled the lightevoked one in time course and magnitude. That the low [Cl"]f ; -induced [TMA + ]O decrease could be 70% to 80% inhibited by retinal bumetanide and 50% to 60% suppressed by choroidal DIDS exposure supports our hypothesis that modulation of RPE Na-K-Cl cotransport can modify subretinal volume. We have not yet investigated the effect of changing subretinal [Na + ]o. We would expect subretinal [Na + ]o to rise during illumination because of closure of the light-sensitive channels of the photoreceptors, and this should stimulate, not suppress, Na-K-Cl cotransport. We also have not measured subretinal [Cl~]o during illumination, but with [K + ]o falling and [Na + ]o presumably increasing in the space during illumination, [Cl"]o may not change. We do not have evidence to support K + as the only extracellular ion driving the volume increase, but we consider it likely that the prominent light-evoked subretinal [K + ]o decrease is a key part of the mechanism.

In addition, the physiological and pharmacokinetic roles of human MCT6 remain to be elucidated. The uptake of [3H]probenecid by MCT6-expressing oocytes at pH 6.0 was lower than that by control oocytes. Further investigation is necessary to compare the properties of probenecid uptake by MCT6 with those of bumetanide uptake and to understand the action of pH on the transport activity of MCT6. MCT6 did not transport PAH and ES, typical substrates of organic anion transporters such as OATs and OATPs, which are involved in.

Inhibitors. In the first series of experiments, we examined the effects of protein phosphatase inhibitors on bumetanide-sensitive transepithelial 86 Rb flux in primary cultures of human tracheal epithelial cells grown on permeable supports. Two agents tested were okadaic acid OA ; and calyculin, which are potent inhibitors of serinethreonine specific protein phosphatase 1 and 2A PP1 and PP2A ; . The order of potency for OA is PP2A PP1 Ki of 0.1 nM vs 10 and for calyculin is ten fold higher for PP1 compared to PP2A 15 ; . Pretreatment of serum-deprived human tracheal epithelial cells with 0.1 nM OA caused a 3.5-fold increase in bumetanide-sensitive 86 Rb cumulative flux and a 3.2-fold increase in the percent of total flux that was bumetanide sensitive Table 1 ; , indicating activation of NKCC1. Higher concentrations of OA, up to 10 nM, each significantly increased NKCC1 activity and percent of total bumetanide sensitive flux. In contrast, pretreatment with calyculin at 10 nM, a concentration that blocks PP1 but not PP2A, did not alter NKCC1 function. These results indicate a preferential role for PP2A in the regulation of airway epithelial NKCC1 function. Because of limited availability of human tracheal tissue as a source of epithelial cells, we selected an airway epithelial Calu-3 cell line for further study because this cell line expresses abundant NKCC1 protein and CFTR protein and displays highly regulated transport function 3, 22 ; . As shown in Table 1, pretreatment of Calu-3 cells with 10 nM OA increased bumetanide-sensitive 86 Rb transepithelial basolateral-to-apical flux 4.1 fold and the percent of total bumetanide-sensitive flux 2.9 fold, indicating activation of NKCC1. We also quantitated NKCC1 activity as bumetanide-sensitive 86Rb uptake in Calu-3 cells grown on Transwell filter inserts. As shown in Fig. 1A, OA at a final concentration of 1 nM significantly increased bumetanide-sensitive 86Rb uptake from 21.0 3.8 n 6 ; nmol K mg protein to 37.4 4.3 n 3 ; nmol K mg protein P .02 ; . As a control, cells were treated with methoxamine, a 1-adrenergic agent which stimulates NKCC1 activity in human tracheal epithelial cells and Calu-3 cells. Bumetanide-sensitive 86Rb uptake increased 3.6 fold to 75.5 14.2 n 5 ; nmol K mg protein. Pretreatment with 1 nM OA before basolateral addition of methoxamine further increased the response to methoxamine. In the and byetta.

100.4 million and 7.5 million at December 31, 2005 and December 31, 2004, respectively. These liabilities are undiscounted, do not consider potential recoveries from insurers or other parties and will be paid out over the periods of remediation for the applicable sites, which are expected to occur primarily over the next 15 years. Although it is not possible to predict with certainty the outcome of these matters, or the ultimate costs of remediation, management does not believe that any reasonably possible expenditures that may be incurred in excess of the liabilities accrued should exceed .0 million in the aggregate. Management also does not believe that these expenditures should result in a material adverse effect on the Company's financial position, results of operations, liquidity or capital resources for any year. Pensions and Other Postretirement Benefit Plans Net pension and other postretirement benefit cost totaled 1.8 million in 2005 and 1.5 million in 2004. Pension and other postretirement benefit plan information for financial reporting purposes is calculated using actuarial assumptions including a discount rate for plan benefit obligations and an expected rate of return on plan assets. The Company reassesses its benefit plan assumptions on a regular basis. For both the pension and other postretirement benefit plans, the discount rate is evaluated annually and modified to reflect the prevailing market rate at December 31 of a portfolio of high-quality fixed-income debt instruments that would provide the future cash flows needed to pay the benefits included in the benefit obligation as they come due. At December 31, 2005, the Company changed its discount rate to 5.75% from 6.0% for its U.S. pension plan. The discount rate for the Company's U.S. other postretirement benefit plan remained the same at 5.75%. The expected rate of return for both the pension and other postretirement benefit plans represents the average rate of return to be earned on plan assets over the period the benefits included in the benefit obligation are to be paid. In developing the expected rate of return, the Company considers long-term compound annualized returns of historical market data as well as actual returns on the Company's plan assets and applies adjustments that reflect more recent capital market experience. Using this reference information, the Company develops forward-looking return expectations for each asset category and a weighted average expected long-term rate of return for a targeted portfolio allocated across these investment categories. The expected portfolio performance reflects the contribution of active management as appropriate. As a result of this analysis, for 2006, the Company's expected rate of return of 8.75% remained unchanged from 2005 for its U.S. pension and other postretirement benefit plans. The target investment portfolio of the Company's U.S. pension and other postretirement benefit plans is allocated 45% to 60% in U.S. equities, 20% to 30% in international equities, 15% to 25% in fixed-income investments and up to 8% in cash and other investments. The portfolio's equity weighting is consistent with the long-term nature of the plans' benefit obligation. The expected annual standard deviation of returns of the target and bumetanide.
