Temperature The water quality varied from month to month during the study period. Because carbenicillin is less active at an acid pH, one might think that the atmosphere of 90% CO2 and 10% H2 used in our study might cause some acid inactivation of the antibiotic. However, the pH of the media after incubation was found to be 7.0. Zone sizes were difficult to measure with both disk diffusion methods. There was often an inner zone of growth within the zone of inhibition in the pour plate method. In the surface disk diffusion method the zones were seldom clearly demarcated, and it was not easy to determine the actual zone size. The best regression curve was obtained with the 50-, ug carbenicillin disk and the surface disk diffusion method, although even this curve would not be satisfactory for use in a disk diffusion method. Most strains of Clostridium perfringens were aberrant in their position on the curve, which was undoubtedly due to the rapid growth of this species, resulting in smaller zone sizes in relation to its MIC. Separate curves would have to be used for organisms with differing growth rates in order for the disk diffusion method to be accurate. This is in agreement with the work of Sutter et al. 7 ; , who have shown that different zone sizes must be used to indicate susceptibility of different anaerobes to the various antibiotics. Likewise, Riff and Jackson 22 ; noted a reduction of approximately 60% in the tl2 of gentamicin among four patients to whom carbenicillin and gentamicin were administered concomitantly in a dosage ratio of 80: 1. In our studies, maintenance of serum ticarcillin levels within a range considered to be "therapeutic" 56 to 169 ug ml ; resulted in considerably less inactivation of gentamicin in vitro and in vivo than observed with a therapeutic range of carbenicillin 107 to 308 jig ml ; . Although the reduction of gentamicin t112 by ticarcillin in patients with renal failure was statistically significant, the actual mean reduction was only 26%. Furthermore, there was essentially no change in the t1 2 of gentamicin among three of eight patients given ticarcillin. Since it is likely that ticarcillin will be administered in dosages that are 30 to 50% lower than the recommended dosages of carbenicillin 8 ; , the degree of gentamicin inactivation by this antibiotic in clinical use will probably be less than that experienced with carbenicillin.
