Methods: Male Rattus norvegicus were divided into 2 groups: normothermic C, ambient temperature 24C ; and heat acclimated AC, ambient temperature 33C1 ; . Animals from those groups were subjected to 2 hours of heat stress at 41C1 and sacrificed following different recovery periods at normothermic conditions. Protein levels from mitochondrial and cytosolic cellular fractions from rat hearts were evaluated by Western blot technique. mRNA levels were estimated by using RT-PCR and verified by qRT-PCR. Results: Following heat stress there were significantly lower levels of Bad mRNA pro-apoptotic member of the Bcl-2 family ; in the AC vs the C animals. 1 hr post heat stress, the BclXL anti-apoptotic ; Bad proteins ratio in the mitochondrial fraction was 2.470.2 and 1.40.4 for AC vs C hearts, respectively. Likewise, mitochondrial cytosolic cytochrome C ratio was 2.070.18 and 0.940.22 for the AC and the C groups. Significantly lower protein levels of caspase 9 and of the active form of caspase 3 were found in the AC compare to the normothermic animals. In contrast, no change in caspase 8 transcripts an indicative of FAS ligand apoptotic pathway ; in both groups was observed. Acclimated hearts responded faster to the heat stress. Conclusions: Our data indicates that heat acclimation enhances anti-apoptotic pathways in response to heat stress via shifting the balance between pro- and antiapoptotic Bcl-2 proteins, thus implying mitochondrial mediated cytoprotective activity. In contrast, no indication for alteration in the FAS ligand was observed and dolasetron.
