An investigation aimed at optimizing the yield of A2E Fig. 1 ; from all-trans-retinal and ethanolamine was performed by using a wide range of reaction conditions. Typical results are summarized in Table 1. When this reaction was carried out in the absence of acetic acid, only a trace amount of A2E was observed after 1 week run 1 ; , whereas conditions using ethanolamine hydrochloride in place of the free amine gave no A2E run 2 ; . The previously used biomimetic conditions.
Tadpoles which had 3.75 ng of l-thyroxine in 50 nl of oil injected into one eye. The projections made by the thyroxine-treated eye to the ipsilateral RVN A ; , NB B ; , CGT C ; , and UF D ; are illustrated. With the occasional exception of the NB, none of these terminal zones normally are innervated at stage 54. Reaction product in the RVN A ; is in the usual lateroventral region of central sections of the nucleus arrow ; . The projection to the NB 6 ; is much more dense and extensive than that typical of PTU-reared tadpoles at stage 54 Fig. l ; , and is subdivided into dense and sparse subregions arrows ; , as is the ipsilateral NB terminal field in normal metamorphosing animals, beginning at stage 60. Density of innervation in the case illustrated is comparable to that seen normally in early postmetamorphic animals. The projections to the CGT C, arrowheads ; and UF D, between arrowheads ; , appear equivalent to that of a normal frog. The ipsilateral projections formed by the untreated eye of a similar tadpole, in contrast, are at the sparse level characteristk of some PTU-reared or nonal tadpoles at stage 54 see Fig. 1 ; . The RVN E ; , CGT G ; , and UF H ; are devoid of reaction product. The NB F ; contains a small amount of reaction product arrowhead ; at the lateral edge of nucleus posterocentralis PC ; . Scale bar 50 pm.
When looking in the mirror, i not concerned, somewhat concerned, or very concerned about the appearance of my wrinkles.
Activated NF- B transcription factor was composed of heterodimers of p50 p65. However, in addition to the p50 p65 heterodimers, L-PAM treatment activated NF- B transcription factors composed of homodimers of p65 and conceivably also heterodimers of p65 Rel B that bound specifically to the NF- B binding site in the enhancer of the human B7-1 gene, which were not seen in the study by Zhao et al. 25 ; . At present the function of the different dimers of the activated NF- B that bind specifically to the NF- B binding site in the enhancer of the B7-1 gene is not known. Consequently, it is not known what the significance is of the fact that in our studies, as in the studies by Zhao et al. 25 ; , one of the activated transcription factors that bound specifically to the NF- B binding site in the enhancer of the human B7-1 gene consisted of heterodimers of p50 p65, whereas the other activated transcription factors were different in the two studies. Finally, it is not known at present whether the differences seen in the composition of some of the activated NF- B transcription factors that bound specifically to the NF- B binding site in the enhancer of the human B7-1 gene in the two studies are the result of differences between the type of cell used i.e., B7-1-expressing mature B cells vs mast cells ; or the result of the "modalities" used to "induce" B7-1 expression i.e., Raji cells that constitutively express B7-1 vs B7-1-negative P815 tumor cells that were induced to express B7-1 by exposure to L-PAM ; . In the current studies we used a cell-permeable peptide that was previously shown to inhibit the binding of NEMO to the IKK complex which consists of IKK and IKK ; , thereby inhibiting IKK phosphorylation of the I B and I B , which in turn inhibits the degradation of the I B that is required for the translocation of NF- B from the cytoplasm to the nucleus where it activates the specific cellular genes 22, 54 ; . Our studies revealed that this peptide, which was previously shown to inhibit cytokine-induced NF- B activation 22 ; , also inhibits L-PAM-induced NF- B activation. Moreover, our studies illustrated that this peptide inhibits L-PAM-induced B7-1 gene expression, indicating that NF- B activation is essential for L-PAM-induced B7-1 gene expression. It is interesting to point out at this stage that the NF- B binding site in the enhancer of the IFN- gene, which is important for IFN- gene expression 55, 56 ; , is identical in sequence to the sequence of the NF- B binding site in the enhancer of the B7-1 gene 25, 53 ; . Thus, although other transcription factors in addition to NF- B are involved in the up-regulation of IFN- gene expression 55, 56 ; , these observations prompted us to determine whether exposure of tumor cells to L-PAM in vitro also leads to up-regulation of IFN- gene expression. Our studies revealed that within 2 h after in vitro exposure of tumor cells to L-PAM there is a massive accumulation of IFN- mRNA, and this accumulation requires de novo RNA synthesis, indicating that L-PAM indeed leads to activation of IFN- gene expression V. Jovasevic and M.B. Mokyr, manuscript in preparation ; . IFN- not just oxidative stress ; may in turn function as an inducible "danger" signal for the immune system 57.

Ala substitution, by contrast, resulted in a markedly decreased ability to bind ThDP and an increased Km for cofactor binding. The Tyr 113-to-Ala substitution resulted in a 50 100-fold increase in the Km for ThDP, indicating that Tyr 113 is indeed important for ThDP binding 10 ; . In 2-oxoisovalerate dehydrogenase from P. putida, Tyr 88, similar to Tyr 102 in the human enzyme, is stacked against the 4 -aminopyrimidine ring with other hydrophobic residues 12 ; . Tyr 133 in the P. putida enzyme has a role similar to that of Tyr 113 in the human enzyme, contributing to the binding of the cofactor through a hydrogen bond with a phosphate oxygen. In the crystal structure of E. coli E1, Phe602 is stacked against the 4 -aminopyrimidine ring, 2 with a role analogous to that of Tyr 88 and Tyr 102 in the two 2 E1 subunits. In summary, Tyr177 in the E. coli E1 enzyme has a different role than the tyrosine residues reported at the active center of the two published structures of the branched-chain pyruvate dehydrogenases with different quaternary structures. In conclusion, the combination of the x-ray structural evidence on the location of Tyr177 with the kinetic and spectroscopic data suggests that Tyr177 has a role in stabilizing some transition state s ; in the reaction pathway, starting with the free enzyme and culminating with the first irreversible step, decarboxylation kcat Km-type effects ; , as well as in some step in reductive acetylation of lipoyl-E2. The results with the Y177F variant suggest that without the hydrogen-bonding ability of the tyrosine, the bulky side chain at position 177 is detrimental to catalysis.
