No covalent binding of 2, 5-diClPh-MeSO2 to rat olfactory microsomes was observed, demonstrating that this isomer was not metabolized to a reactive intermediate by microsomal olfactory CYP enzymes. Interestingly, previous tapesection autoradiograms revealed a high and selective binding of the 2, 5-chlorinated isomer in the olfactory mucosa of mice 2 ; . The uptake of the 2, 5-chlorinated isomer in the mouse olfactory mucosa was comparable to that of the toxic 2, 6chlorinated isomer and did not therefore explain the strikingly different toxicity of the two isomers. The identity of the bound radioactive substance at this site in vivo has not been determined. The lack of covalent binding of 2, 5-diClPh-MeSO2 to olfactory microsomes implies a different binding mechanism for the 2, 5-chlorinated isomer in the olfactory mucosa following in vivo exposure. Several 4-methylsulphonyl-PCBs with chlorine substituents in 2, 5-position bind reversibly to specific ligand-binding proteins in rodent airways and kidney 9, 23 ; . Conceivably, the selective binding of 2, 5-diClPhMeSO2 reported in the olfactory mucosa of mice in vivo could be due to an association of this isomer with a ligand-binding protein residing at this site 12 ; . In conclusion, the lipophilic MeSO2 -derivative 2, 6diClPh-MeSO2 -induced necrosis in the dorsomedial part of the rat olfactory region whereas the 2, 5-chlorinated isomer only induced minor changes at this site in rats. The present study revealed a strikingly higher metabolic activation and covalent binding of 2, 6-diClPh-MeSO2 to rat olfactory microsomes than of the 2, 5-isomer. A CYP2A-dependent biotransformation of 2, 6-diClPh-MeSO2 to a reactive, electrophilic intermediate and irreversible protein adduct formation is suggested to underlie the high toxicity of 2, 6-diClPhMeSO2 in the olfactory mucosa of rats and mice. Covalent binding to critical proteins with loss of function is conceived as the mode of toxic action of a number of drugs and chemicals. The identification and characterization of proteins that are targeted by 2, 6-diClPh-MeSO2 would help understanding the high toxicity of this isomer in the olfactory mucosa.
