Plasmodium falciparum Carrier Recruitment. P. falciparum gametocyte carriers were recruited from five to eleven-year old children in the School district of Mfou, 30 km from Yaounde, Cameroon. Surveys were carried out in collaboration with the medical team in charge of malaria treatment at the local hospital. Thick blood smears were taken from each volunteer, air-dried, Giemsa-stained, and examined by microscopy for the presence of P. falciparum. Children with asexual parasitemia 1, 000 parasites per microliter ; were treated in accordance with national guidelines. Asymptomatic gametocyte-positive children were recruited for the study. We are performing a comprehensive investigation on blood samples from smokers and non-smokers. Because tobacco smoke is a well-documented source of a variety of potentially mutagenic and carcinogenic compounds, smokers should be a suitable study group with relevant mutagen exposure. Here we report our results for a group of 40 smokers and 40 non-smokers. Baseline DNA effect were determined in the absence and the presence of a repair inhibitor aphidicolin ; . Mutagen sensitivity towards benzpyrenediolepoxide BPDE ; and gammaradiation and the repair of DNA damage induced by these two mutagens was comparatively investigated. None of these approaches revealed significant differences between smokers and non-smokers. These results indicate limitations of the comet assay in the detection of smoking-related genetic effects in peripheral blood cells and methotrexate.
