For discovery of new more efficient enzymes or engineering of known ones Schulein, 2000 ; . We here present ciPCR as an alternative to the otherwise well-established cultivation-based enzyme discovery strategy. By looking for new enzymes in the fungal communities carrying out the cellulose turnover in ecosystems, genes originating from poorly culturable and less dominant microorganisms could be recovered Hugenholtz and Pace, 1996; Dawson and Pace, 2002 ; potentially leading to discovery of enzymes with higher substrate affinity and specific activity. In the present study, degenerate primers were developed in an attempt to amplify fungal genes coding for cellobiohydrolase GH7 and endoglucanase GH45 Henrissat, 1991 ; . Composite DNA was extracted from corn stover and leaves which were left on the ground over winter. In addition, fungalspecific primers were used in an attempt to identify the diversity of these filamentous fungi.

Animal Management and Treatments. On May 17, 1982, 98 purebred Hampshire and Suffolk ewes were weighed and randomly assigned within age and breed to one of two treatment groups; supplemented n 50 ; or unsupplemented n 48 ; . Both groups were grazed together on available pastures. Daily at 0900, supplemented ewes were sorted from unsupplemented ewes, offered .45 kg of whole corn grain head, and then returned to the pasture. At least two vasectomized rams, androgenized ewes Scheffrahn et al., 1982 ; , or a combination of these, were with the ewes to detect estrus. Briskets of these animals were smeared with a mixture of paint and grease. The color was changed every 14 d, and ewes were inspected daily for marks indicating estrus. The day when a ewe was marked on or after August 15 ; was designated d O. On ewes began one o f three pre-mating treatments: 1 g of phenobarbital in a gelatin capsule administered orally with a dosing forceps n 31 .45 kg of an 18% crude protein ground grain mixture, consisting of 338 g corn plus 112 g soybean oil meal n 33 or treatment n 34 ; . These three pre-mating treatments had been assigned at random at the initiation of the experiment within age-breedsummer supplementation treatment group subclass. During the pre-mating period, there were six treatment combinations; two summer supplementation treatments in combination with three pre-mating treatments. Treatments were administered daily to each ewe and continued until the ewe was detected in estrus. She was then removed from the treatments, placed with a single intact ram for 24 h, and then p u t lush pasture with no grain supplementation. All treatments and matings stopped on October 7. Immediately prior to exposure to an intact ram, ewes were scored for body condition by three people, independently. The mean o f these scores was the ewe's breeding condition score. The scoring system used was that of Russel et al. 1969 ; , ranging from 1 very thin with increments of .5, to 5 very fat. Six to 13 d after a ewe was placed with an intact ram, corpora lutea were counted at laparoscopy by the procedure of Lamberson and Thomas 1982 ; . Ewes were tranquilized prior and phenylpropanolamine.
FIG. 2. Clustering diagram of samples in the study. The algorithm Eisen et al., 1998 ; was used to cluster the gene expression profiles for known and coded compounds for the set of derived discriminatory genes used in this study. The diagram illustrates that all of the compounds separate into 2 distinct nodes. Node I represents samples that are related to phenobarbital and Node II represents samples classified as peroxisome proliferators. Red indicates genes that are induced by treatment and green indicates repression of expression by treatment.

Saturation of the enzyme responsible for demethylation of the parent compound or inhibition of this enzyme by the demethylated product. At the time of admission, the patient's serum concentration of primidone was 62 mg liter, six-fold the upper limit of the therapeutic range for this drug 2 ; . Thereafter the primidone concentration increased to a maximum of 130 mg liter at 60 h. This time course is most compatible with continued absorption of the drug from the gut. The presence of phenobarbital in our patient's serum could bedue either to formation of this compound in vivo from primidone, or to ingestion of the barbiturate directly: The time course of appearance of phenobarbital in her serum Figure 5 ; is compatible with continued absorption of its metabolic precursor, primidone, as well as with sustained absorption of the barbiturate itself. Because of the high plasma concentrations of methsuximide, primidone, and their active metabolites, it was not possible to determine the contribution of the individual compounds to the profound coma seen in this patient. After acute overdosage with the drugs this patient took, they would be cleared from the body only by metabolism. Under such conditions in the patient reported by Brillman et al. 5 ; , the plasma half-life of phe# obarbital observed in primidone overdosage was 4.5 days, as compared to 15 h for the parent compound. In our patient the concentrations of phenobarbital and primidone declined to about half their peak values by 60 h and 30 h, respectively. We have not calculated halflives for the clearance of these drugs from serum because of concurrent removal of the compounds by peritoneal and hemodialysis Figure 5 ; . Peritoneal fluid obtained before the 60-h peritoneal dialysis was begun contained 225 mg of N-desmethylmethsuximide per liter, while serum obtained concurrently contained 278 mg of this drug per liter, which indicates that N-desmethylmethsuximide can cross the peritoneal membrane. This observation prompted the clinical staff to institute and sustain peritoneal dialysis. During the first 2 h of pentoneal dialysis, 280 mg of N-desmethylmethsuximide was removed. When the patient's blood pressure stabilized, she was also treated by hemodialysis. During a 2.5-h period ofthe first hemodialysis, approximately 1 g of N-desmethylmethsuximide accumulated in the dialyzer bath. As shown in Figure 5, after 216 h of treatment all drugs in her serum had decreased to either subtherapeutic or nonmeasurable concentrations. Further information on some components. We thought that the unidentified peak observed in the routine gas-chromatographic analysis of the serum sample Figure 4 ; might be another metabolite of methsuximide and therefore we conducted a more detailed investigation by use of combined gas chromatography-mass spectrometry GC MS ; . Three samples of serum were chosen for analysis by GC MS: one obtained at the time of admission, a second 24 h after admission, and the third at 36 h. Ethyl acetate extracts of these sera were injected on column without derivati and pilocarpine.

Radiologic remodeling phenomena, such as cancellous densification and cortical thickening, around a hip prosthesis have various incidences in the literature, even when studied with the same prosthesis. Also relations between these features and clinical items, such as pain and limping, are described. We prospectively studied the clinical and radiologic results in 3 patient groups: 89 standard primary THRs, 63 primary THRs in patients with hip dysplasia, and 48 revision. THRs. We describe the differences in clinical behavior of a titanium HA-coated stem with variations in tightness of the fit of the prosthesis in the femoral canal, operative. group, stem size, grade of osteoporosis and femoral morphometry. The clinical results in all three groups are slightly favorable for the standard primary group. The pain relief and stem survival in the three groups were similar to that reported in other studies regarding cementless prostheses. Nontight femoral fit significantly positively influenced the development of cancellous densification; however the presence. of osteoporosis also contributed to the formation of cancellous densification. Only tight femoral canal fit significantly positively influenced the development of cortical thickening independent of other parameters. There was no correlation between clinical parameters and the radiologic phenomena in either of the groups. O-274 and pima. A number of medications similar to phenobarbital were developed, including primidone.
Drug Interactions Care should be taken using Sandimmun&# cyclosporine ; with nephrotoxic drugs since potential synergies of nephrotoxtcity may occur Concurrent administration of rifampin. phenytoin, phenobarbital and I V sulfatrimethoprim may decrease plasma levels of cyclosporine Cimetidine, ketoconazole. and amphotericin B may increase the plasma concentration of cyclosporine Monitoring of plasma or blood levels of cyclosporine are essential when these drugs are used concomitantly Carcieogeeeois.
